Journal of Experimental Medicine is a monthly peer-reviewed medical journal published by Rockefeller University Press that publishes research papers and commentaries on the physiological , pathological , and molecular mechanisms that encompass the host response to disease. The journal prioritizes studies on intact organisms and has made a commitment to publishing studies on human subjects. The journal has no single editor-in-chief , but thirteen academic editors. Welch , the school's founder and also the first president of the Board of Scientific Directors of the Rockefeller Institute since renamed Rockefeller University. From its inception, Welch edited the journal by himself—even editing manuscripts while attending baseball games. By March , the editorial burden became too great for Welch, who stopped publishing papers and began stockpiling manuscripts and unanswered correspondence in his office, explaining the conspicuous absence of published papers from to In October , Welch appealed to the board of the Rockefeller Institute to take over the journal. The transfer of ownership and publication responsibilities required the physical transfer of manuscripts from Welch's office, which fell to the director of the Rockefeller Institute, Simon Flexner , who carried the abandoned manuscripts from Baltimore to New York City in a suitcase. The first issue published by the Rockefeller Institute appeared in February , with Flexner serving as editor, and the journal has been published regularly since then. Although the journal was adopted by the Rockefeller Institute as a venue for publication of the institute's own research, it also accepted submissions from outside. Even in the early years, more than half of the papers published in the journal came from external labs.

The intestine plays an important role in nutrient digestion and absorption, microbe defense, and hormone secretion. Although major cell types have been identified in the mouse intestinal epithelium, cell type-specific markers and functional assignments are largely unavailable for human intestine. Here, our single-cell RNA-seq analyses of 14, epithelial cells from human ileum, colon, and rectum reveal different nutrient absorption preferences in the small and large intestine, suggest the existence of Paneth-like cells in the large intestine, and identify potential new marker genes for human transient-amplifying cells and goblet cells. We have validated some of these insights by quantitative PCR, immunofluorescence, and functional analyses. Furthermore, we show both common and differential features of the cellular landscapes between the human and mouse ilea. Therefore, our data provide the basis for detailed characterization of human intestine cell constitution and functions, which would be helpful for a better understanding of human intestine disorders, such as inflammatory bowel disease and intestinal tumorigenesis. Over the past decade, our view of human-associated microbes has expanded beyond that of a few species toward an appreciation of the diverse and niche-specialized microbial communities that develop in the human host with chronological age. The largest reservoir of microbes exists in the distal gastrointestinal tract, both in the lumen, where microbes facilitate primary and secondary metabolism, and on mucosal surfaces, where they interact with host immune cell populations. While local microbial-driven immunomodulation in the gut is well described, more recent studies have demonstrated a role for the gut microbiome in influencing remote organs and mucosal and hematopoietic immune function. Unsurprisingly, therefore, perturbation to the composition and function of the gut microbiota has been associated with chronic diseases ranging from gastrointestinal inflammatory and metabolic conditions to neurological, cardiovascular, and respiratory illnesses. Considerable effort is currently focused on understanding the natural history of microbiome development in humans in the context of health outcomes, in parallel with improving our knowledge of microbiome-host molecular interactions. These efforts ultimately aim to develop effective approaches to rehabilitate perturbed human microbial ecosystems as a means to restore health or prevent disease. This review details the role of the gut microbiome in modulating host health with a focus on immunomodulation and discusses strategies for manipulating the gut microbiome for the management or prevention of chronic inflammatory conditions.
