That choice can go as far as the final point before the end of the story. While I try to keep it as a minimum where possible, this post will contain light spoilers for the end of Cyberpunk Before you complete the game, be sure to get some cyberpunk point of no return you might need.
This point becomes available as soon as you complete Tapeworm ; however, it's highly recommended to complete the questlines for Johnny and Kerry first. Enter the elevator of Embers and head up to the thirteenth floor to talk with Hanako Arasaka there. She will state that her brother has to answer for his patricide and divulge the location of Mikoshi, the only place where you and Johnny can be separated from each other: beneath Arasaka Tower. She suggests that you help her expose Yorinobu Arasaka as the murderer of their father, Saburo, to the Arasaka board of directors and in exchange she will help you get rid of Johnny's construct in your head. Return to the elevator and Johnny will suggest that he takes control of your body to raid Arasaka Tower with Rogue instead, before you black out due to a Relic malfunction. You wake up in Viktor's clinic, where he will give the prognosis that the next Relic malfunction will likely kill you. Take the last of the pills on the tray and head outside, where Misty will suggest going up to the roof to decide what's next.

It doesn't really lock you out: once you talk to her you are committed till you end the game, however no matter what ending you choose there is going to be option "Just one more gig" and you are going to be thrown back to a point where you are standing before elevator and you are free to do all stuff you want and replay the ending again. However all loot and levels you gather during the ending mission are wiped.
