Amado, a principal member of Kara who oversaw their research and development, kidnaps Shikadai and seeks asylum in the Leaf Village. In exchange, he promises to tell them everything he knows. Although he is someone they cannot trust, with Shikadai's life at stake the Hokage personally gets involved in the negotiation. In exchange for being granted asylum in the Village Hidden in the Leaves, Amado reveals everything he knows. Amado and Koji Kashin have been working together to bring down Jigen. He tells everyone that Jigen's true identity is Isshiki Otsutsuki, who accompanied Kaguya Otsutsuki to their world ages ago. He goes on to explain the Karma, the mark that plagues Boruto and Kawaki. If Jigen regains the power of the Otsutsuki Clan, it will be impossible to counter him. Before that happens, Kashin tries to drag out Isshiki, who has taken over Jigen's body using a barrage of carefully devised techniques in order to destroy him. Through the tool that Amado uses, Naruto and the others are able to watch the battle from Leaf Village. Finally, Koji's plan works and their strongest and most evil foe, Isshiki Otsutsuki, appears!

The Otsutsuki Clan is the strongest of all clans in the Naruto series. In time Kaguya gave birth to twin sons: Hagoromo and Hamura , both of whom were born with chakra of their own. Afterwards, Kaguya eventually found herself ruled by her own desire to possess all chakra, in which made her turn against her own sons. Having devised a plan to absorb their chakra, she integrated with the Earth's God Tree and became the Ten-Tails. The Ten-Tails' chakra was sealed into Hagoromo while its husk, the Demonic Statue of the Outer Path , was hidden away and sealed on the Moon, in order to prevent the Ten-Tails' revival. Before being sealed, Kaguya created a parasitic lifeform containing her will, Black Zetsu to find a way to revive her.

Many thought this alien species was out of the picture after Kaguya, Kinshiki and Momoshiki died, but now it's been revealed the lattermost of those marked Boruto as a vessel too as part of a major return for the clan. Jigen was also revealed to have a Ten Tails, the same sort of creature that helped Kaguya almost end the world in the Naruto finale. This causes things to go from bad to worse, and it won't be getting any better, as it seems the villain might have access to more than one. In the Naruto series, chakra were the life essence of humanity, with Kaguya's teaching her sons, Hagoromo and Hamura, to harness nature and help spread this latent energy to humans. But it turns out, she grew bitter and wanted to absorb back this chakra, using the Ten Tails who'd then turn into the God Tree, trap everyone in its branches and feed off the chakra. This created a chakra fruit, which Kaguya and her kind would eat. While at first it seemed to be for sustenance and fun, it turns out it's part of a bigger scheme: evolution. The aliens actually seed out these Ten Tails beasts all across the cosmos as scouts that pick planets with unique creatures to feed off. They suck the citizens dry, no matter what species they are, to enhance the fruit.

It would also cause a metamorphosis to his being, increasing his size and muscular tissue.
